I live in the US and we have a ton of foreign exchange students that come and work for the summer where I live. A lot of them end up staying. I'm friends with a gentlemen from Russia he's been here going on five years. They never lose their accent it more or less becomes less thick.
You can gain any accent. It's possible that you never lose your accent, but you will begin to sound more American. Some people pick up different accents fairly easily.
I am not sure that there is an American accent as there are so many different accents throughout the country that it would be hard to label one American. There are parts of the country where I have trouble understanding people when they speak with the particular local accent. I think when people relocate somewhere long term, there is a tendency to pick up certain phrases and parts of speech, and their way of speaking may change a little as a result. But I don't think most people fully adopt the local accent of a new area.
Of course you can just pull a Hugh Laurie and fake it.

I have a friend who moved to the United States almost 10 year ago and the first thing I noticed is that the he would utilise a type of American accent is with terminology or Americanisms that we don't use here in New Zealand. His accent to a large extent has remained the same.
