I was afraid of my bedroom growing up and well into my teens. It was just something about that room I couldn't stand. I slept in my dad's bed until I was about 9, then on the couch in my brother's room until he kicked me out. Then I spent a few years on the livingroom couch. No fear of the dark or of sleeping alone. Just of that room. There are many theories as to why a particular room can make a person uncomfortable. In your brother's case it may not be a fear so much as a feeling of unease that keeps him from getting a good sleep.
