For such a serious subject, I'm a little surprised it's only come up twice in a 4 year relationship. Your boyfriend was certainly presumptuous in thinking you were joking about such a thing, but it's also possible you hadn't been clear enough about. I don't know either of you at all to be sure. I don't think there's much you can do to prevent the breakup aside from conceding to have children, but I hope that you don't. A breakup over this thing is certainly not unusual, though I'm sure with how long the relationship has been it will be far rougher than if had been addressed properly the first time.
